Migration of Myogenic Cells in the Developing Limb
نویسنده
چکیده
Myogenic cells migrate into the limb from the somitic mesoderm, taking up positions in the dorsal and ventral compartments before differentiating into the limb musculature. This process is influenced by the tissues of the limb in a number of ways. Cells of the lateral part of the somitic dermomyotome are induced to express a migratory myogenic phenotype by a diffusible signal originating in the limb bud mesoderm. The limb and body wall musculature are entirely derived from cells of this migratory myogenic lineage. Components of the extracellular matrix of the limb mesoderm, such as fibronectin, facilitate myogenic cell migration by providing sites for adhesion and directed migration. Differences in the composition of the matrix along the proximo-distal axis make the distal mesoderm more attractive for migrating myogenic cells. Anterior-posterior movement of myogenic cells within the limb bud is highly restricted, resulting in a regionalised contribution of cells to the limb musculature based on somitic origin. Lastly, there is a process that excludes myogenic cells from the most distal region of the limb bud mesoderm; a similar process is involved in the aggregation of myogenic cells into the dorsal and ventral premuscle masses. These findings illustrate the high degree of control over myogenic cell migration mediated by both diffusible factors and the extracellular matrix in the developing limb mesenchyme.

Fragments of quail wing bud containing myogenic cells of somitic origin and fragments of quail sphlanchopleural tissue were introduced into the interior of the wing bud of fowl embryo hosts. No movement of graft into host tissue occurred in the control, but myogenic cells from the quail wing bud fragments underwent long migrations in an apical direction to become incorporated in the developing ...
متن کاملPax-3 is required for the development of limb muscles: a possible role for the migration of dermomyotomal muscle progenitor cells.
Limb muscles in vertebrates originate from dermomyotomal cells, which during early development migrate from the ventrolateral region of somites into the limb buds. These progenitor cells do not express any muscle-specific marker genes or myogenic transcription factors until they reach their destination in the limbs. Here, we demonstrate by in situ hybridization that myogenic cells in somites an...

Retinoic acid (RA) plays an essential role in the development of many embryonic tissues, including the developing tetrapod limb bud. At early stages of limb development, RA levels are highest proximally and regulate the migration of myoblasts into the limb. As the premyogenic progenitor cells migrate into the limb and accumulate in premuscle masses, they express Pax3 and Meox2. Myogenic differe...
متن کاملSF/HGF is a mediator between limb patterning and muscle development.
Scatter factor/hepatocyte growth factor (SF/HGF) is known to be involved in the detachment of myogenic precursor cells from the lateral dermomyotomes and their subsequent migration into the newly formed limb buds. As yet, however, nothing has been known about the role of the persistent expression of SF/HGF in the limb bud mesenchyme during later stages of limb bud development. To test for a pot...
متن کاملLoss of fibroblast growth factor receptors is necessary for terminal differentiation of embryonic limb muscle.
Early in embryogenesis, precursors of the limb musculature are generated in the somite, migrate to the limb buds and undergo terminal differentiation. Although myogenic differentiation in culture is affected by several growth factors including fibroblast growth factor (FGF), it remains uncertain whether migration and differentiation of myogenic cells in vivo are directly regulated by such growt...
